摘要
应用AcA54超凝胶柱层析分离纯化日本血吸虫成虫和虫卵可溶性抗原,分离后的成虫和虫卵可溶性抗原峰型基本相同,但成虫在2个主峰之间比虫卵多1个明显的小峰。成虫第1个峰型的高低与高速离心时的上悬聚集物有关。经ELISA检测柱层析后2种抗原的血清学活性,其分布基本一致。银染色结果表明,此类分离纯化尤适于小于30KD组分的分离,在过柱后可以一次性达到生化纯度。
Soluble antigens from Schistosoma japonicum adult worm and egg (AWA and SEA) were purified by AcA54 ultrogel column choromatography.The protein peaks of choromatography between the two antigen solutions were similar,but an additional small peak was seen between the two major peaks for AWA.The level of the first peak was related to the size of the floating material on the surface of the supernatant after a high-speed centrifugatlon.Serologic antigenic activities of the two antigen solutions after choromatography were not different by ELISA.The result of silver staining of AWA after SDS-PAGE suggested that this method of isolation should be suitable for the purification of less than 30KD components.
