应用小鼠胚胎体外培养技术研究氟的胚胎发育毒性 被引量:1

摘要 目的 探讨无机氟的发育毒性及机理。方法 妊娠 8.5d大鼠胚胎于体外旋转培养系统中给予不同剂量的氟化钠 ,培养 48h后 ,测定卵黄囊直径 ,胚胎体长和头长 ,用Maele Fabry形态学评分系统进行器官形态评分 ,评价胚胎器官分化发育情况。结果 随着氟剂量的增加 ,反映胚胎生长发育和器官分化的各项指标呈现出下降趋势 ,有一定的剂量效应关系。其中 ,卵黄囊及血管网发育分化对氟较敏感 ,剂量为 5 .0mg·L- 1 时与对照比较已有显著性差异。大于或等于 1 0 .0mg·L- 1 时对多数指标影响显著 ,Maele Fabry形态学总评分与对照相比呈现显著差异。胚胎畸形发生率在剂量为 1 5 .0mg·L- 1 明显升高 ,主要表现为神经管闭合不全和体翻转不全。 Objective To understand the developmental toxicity of fluoride.Methods Embryos of Kunxing mouse on day 8.5 of gestation were explanted and cultured in a whole embryo culture system with exposure to F concentrations from 0.0 to 30.0 mg·L -1 for 48 h. After 48 h in culture, each viable embryo was evaluated by Maele Fabry morphological scoring system . Yolk sac diameter, crown rump length, head length and embryonic dry weight were measured. Results Dose dependent decreases in embryonic development parameters and Maele Fabry scores were observed with increasing of F concentration. Yolk sac development scores decreased significantly at 5.0 mg·L -1 When exposure doses at ≥10.0 mg·L -1 , the embryonic development and morphogenetic differentiation were obviously inhibited ,as well as the incidence of dysmorphogenesis significantly increased, including dorsiflexion and neural tube defects. Conclusion The results suggested that aluminum is a developmental toxicant and dysmorphogent in mouse.
