摘要
目的 :探讨非那雄胺对早期糖尿病大鼠视网膜血管内皮生长因子 (VEGF)和微血管密度(MVD)干预的效果。方法 :2 4只早期糖尿病大鼠随机分成A (空白对照组 ) ,B (阿卡波糖 2mg·kg- 1) ,C(阿卡波糖 2mg·kg- 1+非那雄胺 0 .1mg·kg- 1) ,D(阿卡波糖 2mg·kg- 1+非那雄胺 0 .2mg·kg- 1) 4组 ,服药 30d后处死取出视网膜做检查 ,用免疫组织化学检测血管内皮生长因子的表达和微血管密度。结果 :A ,B 2组血管内皮生长因子表达的阳性率高达 10 0 % (12 / 12眼 ) ,C组降至 8% (1/12 ) ,D组为 0。微血管密度各组间比较 ,差异有非常显著意义 (P <0 .0 1)。结论
AIM: To investigate the effects of finasteride on expression of vascular endothelial growth factor (VEGF)and microvascular density (MVD) in retina of early diabetic rats. METHODS: Twenty four early diabetic rats were randomly divided into 4 groups: group A (control group), group B(acarbose 2 mg·kg -1 ),group C(acarbose 2 mg·kg -1 +finasteride 0.1 mg·kg -1 ),and group D(acarbose 2 mg·kg -1 +finasteride 0.2 mg·kg -1 ). After 30 d of drug treatment, the rats were euthanized and their retinas examined. The expression of VEGF and MVD were evaluated by immunohistochemistry. RESULTS: Expression of VEGF was 100 % (12/12 eyes) in both groups A and B, whereas it decreased to 8 %(1/12) in group C, and to 0 in group D. Expression of MVD was very significant difference in the four groups ( P <0.01). CONCLUSION: Finasteride is effective to prevent the VEGF expression and microvascular development in retina of early diabetic rats.
