维生素K_2对HL-60细胞生长及凋亡、分化的诱导作用

摘要 目的 观察维生素K2 (VK2 )对HL 6 0细胞生长及凋亡、分化的诱导作用。方法 通过四氮唑蓝比色 ,细胞形态 ,流式细胞仪测定细胞周期、凋亡率及CD1 4的表达 ,观察VK2 对HL 6 0细胞的影响。结果 HL 6 0细胞经1 0 μmol L以上浓度的VK2 作用后增殖受到抑制 ,1 0 μmol L、2 0 μmol LVK2 作用 72h后 ,细胞形态呈现凋亡细胞的特征。 5 μmol L、1 0 μmol L、2 0 μmol LVK2 作用于HL 6 0细胞 72h凋亡率分别为 7.1 6 %、1 1 .31 %、2 0 .36 % ,与对照组相比差异显著。G0 G1 期细胞阻滞 ,CD1 4的表达与对照组相比无差异性。结论 VK2 对HL 6 0细胞有生长抑制作用 ,能诱导HL 6 0细胞发生凋亡 ,具有一定的量效和时效关系 ,单独使用VK2 的分化作用不显著。 Objective To study the effects of vitamin K 2 on the growth and apoptosis, differentiation of human acute promylocytic leukemia(APL)cell line HL 60.Methods We examined the effects of the growth of cells by MTT test, and observed the changes of morphologic features of cells. Cell cycle, CD 14 expression and rate of apoptosis were detected by a flow cytometry.Results After exposing to above 10 μmol/L vitamin K 2, the proliferation was inhibited.The rates of 5 μmol/L,10 μmol/L and 20 μmol/L VK 2 were 7.16%, 11.31% and 20.36%, which showed statistic difference compared with control group. Exposed to different dose of VK 2 for 72 h, G 0/G 1 cell arrest, but the expression of CD 14 was not notablely enhanced than control group.Conclusion Vitamin K 2 can inhibit the proliferation and induce apoptosis of HL 60 cells, however the differentiation of cells can not be observed by vitamin K 2 alone.
