摘要
目的 观察CIK(cytokine inducedkiller)细胞的体外增殖和杀瘤活性及对Lewis肺癌小鼠脾细胞分泌细胞因子浓度变化的影响。方法 常规方法培养CIK和LAK细胞 ,建立Lewis肺癌小鼠模型 ,给予CIK和LAK细胞治疗 3周后 ,杀鼠取脾 ,用ELISA方法分别测定C5 7小鼠脾细胞上清液的IL 2、IL 4、IFN γ浓度 ,用MTT方法测定C5 7小鼠脾细胞上清液TNF α浓度。结果 CIK治疗的Lewis肺癌小鼠脾细胞培养上清液细胞因子水平明显高于LAK组。结论 CIK细胞对Lewis肺癌小鼠治疗有效 ,杀瘤活性强于LAK ,CIK治疗组荷瘤鼠的细胞因子水平也高于LAK组。
Objective To evaluate the proliferation profile and the cytotoxicity of CIK cells and their anti tumor effects on Lewis mice and to test cytokine of spleen cell medium.Methods CIK and LAK cells were induced by culturing PBMC with regular method. Model of Lewis pulmonary carcinoma was created,and ten CIK and LAK were injected intravenously into Lewis pulmonary carcinoma mice. After 3 weeks mice were killed and spleen cells were cultrued,IL 2, IL 4 and IFN r were tested by ELISA, respectively and TNF α was tested by MTT.Results Level of cytokine in CIK therapy group was higher than that of the LAK.Conclusion CIK cells have high effects on anti Lewis pulmonary carcinoma compared with LAK cells in vivo . The CIK therapy is possibly correlated with induction of the activation of the splenic lymphocytes secreting cytokine.
