GABA对谷氨酸和低糖诱导的人少突胶质细胞瘤细胞凋亡增加的影响(英文)

摘要 目的 观察GABA对谷氨酸和低糖诱导的人少突胶质细胞瘤细胞凋亡增多的影响 ,并探讨其可能的机制。方法 采用免疫组织化学方法和相关的试剂盒测定凋亡百分率的变化和bcl 2、bax、c fos蛋白的表达。结果 ①GABA可以显著抑制谷氨酸和低糖诱导的细胞凋亡百分率的增加 (P <0 .0 5 ) ;②GABA对谷氨酸和低糖诱导的bcl 2、bax蛋白的表达增加无明显影响 ;③谷氨酸 ,低糖和GABA对c fos蛋白的表达没有明显影响。结论 ①GABA可能通过降低细胞凋亡的百分率而具有神经保护作用 ;②GABA的神经保护作用可能是通过其它途径而与bcl 2、bax蛋白的表达无关。 Objective To observe the effect of GABA on increased apoptosis of human oligodendroglioma cell (HOC) induced by glutamate (Glu) and low glucose (LG) and find out the potential mechanism.Methods Immunohistochemical staining methods and the kits concerned were employed to examine changes of apoptotic percentage and expression of bcl 2, bax and c fos proteins.Results ①GABA inhibited the increase of percentage of HOC apoptotic death mediated by Glu and LG ( P <0.05);②GABA had no marked effect on the overexpression of bcl 2 and bax proteins induced by Glu and LG;③After 24h treatment, Glu and LG didn't regulate the expression of c fos protein and GABA also didn't have influence on it.Conclusion ①GABA may have neuroprotection through inhibiting the increased amount of apoptotic cell death mediated by Glu and LG;②The potential mechanism of neuroprotective effect of GABA isn't related to bcl 2 and bax proteins.
