托吡酯单药及合并用药治疗癫痫的临床观察

Topiramate as monotherapy or adjunctive therapy for epilepsy

目的 观察托吡酯单药及合并用药治疗各种类型癫痫的临床疗效、有效剂量及副作用。方法 2 5例癫痫患者行托吡酯单药治疗 ,2 0例癫痫患者行托吡酯添加治疗 ,观察其疗效及副作用。结果 单药治疗组有效率为76.0 %,平均有效维持剂量为 ( 118.1± 5 8.2 )mg·d- 1 ;合并用药组有效率为 70 .0 %,平均有效维持剂量为 ( 14 8.4± 83 .5 )mg·d- 1 ;托吡酯常见的副作用有食欲减退、体重减轻等 ,多发生于加药期 ,症状多轻微且短暂。结论 托吡酯单药及合并用药治疗各种类型癫痫疗效良好。

Objective To evaluate the clinic effect,therapeutic dose and adverse effect of topiramate as monotherapy or adjunctive therapy for different types of epilepsy.Methods 45 patients received topiramate,25 of them as monotherapy,and the other 20 patients as add on therapy.Both the therapeutic and adverse effects were investigated.Results Efficacy in favor of topiramate as monotherapy was 76.0%,with mean sustaining dose at (118.1±58.2)mg·d -1 ,while that as adjunctive therapy was 70.0%,with mean sustaining dose at (148.4±83.5)mg·d -1 .The most common adverse effects were poor appetite and weight loss,which tended to be mild and temporary and often occur when adding the dose.Conclusions Topiramate as either monotherapy or adjunctive therapy is effective and safe for different types of epilepsy.
