黄芪对急性弓形虫RH株感染小鼠的保护作用研究

PROTECTIVE EFFECT OF ASTRAGALUS MEMBRANCE ON MICE OF ACUTE TOXOPLASMOSIS

目的 探讨黄芪颗粒剂对刚地弓形虫 RH株速殖子感染小鼠的治疗作用。方法 ICR小鼠腹腔注射 10 5、10 3、10 2 个速殖子 ,感染后第 1天起每鼠给予黄芪 0 .0 75 g/ d、阿齐霉素 [15 0 mg/(d· kg) ]灌胃治疗 ,对照组给蒸馏水。连续 10 d,观察小鼠的存活率与存活时间。于感染后第 4、8天处死 10 2 个速殖子感染的小鼠 ,PCR荧光定量方法检测肝、肺弓形虫 DNA基因拷贝数。结果 10 5、10 3速殖子感染时 ,黄芪组小鼠平均存活时间为 5 .5 7d和 6 .2 3d,与对照组相比差异无显著性 (P>0 .0 5 ) ,阿齐霉素组小鼠平均存活时间为 6 .96 d和 8.12 d,明显高于对照组 (P<0 .0 5 )。10 2个速殖子感染时 ,黄芪组小鼠平均存活时间为 8.74 d,明显高于对照组 ,阿齐霉素组小鼠平均存活时间为7.79d,与对照组相比差异无显著性 (P>0 .0 5 )。黄芪组小鼠感染 10 2个速殖子后第 4、8天肝、肺虫荷与对照组相比差异无显著性 (P>0 .0 5 )。结论 黄芪可以改善低剂量 (10 2 )

Objective To study the protective effect of Astragalus membrance on mice infected with tachyzoites of Toxoplasma gondii. Methods ICR mice were infected intraperitoneally with 10 5, 10 3, 10 2 tachyzoites of virulent RH strain of Toxoplasma gondii, and the mice were orally treated with Astragalus membrance 0 075 g/d per mouse or Azithromycin [150 mg/(d·kg)] each day starting from day 1 post-infection for 10 days. The survival rate and period were investigated. The parasite loads of livers and lungs of the mice infected with 10 2 tachyzoites were determined by fluorescence PCR methods at 4 day-post-infection (dpi) and 8 dpi. Results When infected with 10 5, 10 3 tachyzoites, treated with Astragalus membrance, the average survived days of the mice were 5 57 days and 6 23 days, and treated with azithromycin were 6 96 days and 8 12 days respectively. The azithromycin group but not the astragli group survived significantly longer than the control(P<0 05). When infected with 10 2 tachyzoites, the mice treated with Astragalus membrance survived 8 74 days, and treated with azithromycin 7 79 days. The astragli group but not azithromycin group survived significantly longer than the control(P<0 05). There was no difference between parasite loads of the control group and astragali group in either livers or lungs at 4 and 8 dpi. Conclusion Astragalus membrance enhanced the survival of mice infected with low dose of RH tachyzoites (10 2).
