反相高效液相色谱法测定盐酸托烷司琼的人体血药浓度 被引量:8

摘要 目的 建立测定盐酸托烷司琼的人体血药浓度的方法 ,用于其血药浓度测定并进行临床药代动力学研究。方法 采用高效液相 -二极管阵列色谱法测定盐酸托烷司琼的血药浓度。结果 盐酸托烷司琼的校正标准曲线方程为 ^Y= - 0 0 35 7+0 0 1 7X(r=0 990 9) ,其 5 ,1 0 ,2 5 μg·L-1 3浓度的血样回收率分别为 1 0 3 2 4 % ,97 5 1 % ,1 0 4 95 % ;精密度分别 5 6 7% ,7 83% ,8 1 8% ;志愿受试者禁食口服 2 0mg盐酸托烷司琼胶囊后 ,盐酸托烷司琼的Cmax为 2 7 4 4 μg·L-1 。结论 此方法可满足测定要求 。 S AIM To develop a method which can determine the blood concentration of tropisetron chlorate in order to study its clinical pharmacokinetics. METHODS The blood concentration of tropisetron chlorate was determinated by HPLC. RESULTS Calibrated standard curve of tropisetron chlorate in blood is =-0 035 7+0 017 X ( r =0 990 9), the recoveries of three concentrations 5, 10, 25 μg·L -1 are 103 24%, 97 51%, 104 95%, respectively; and the precisions are 5 67%, 7 8%,8 18%, respectively. After 20 mg tropisetron chlorate were administered by a voluteer via oral, C max of tropisetron chlorate was 27 44 μg·L -1 . CONCLUSION The method is good enough to study pharmacokinetics of tropisetron chlorate.
