摘要 目的:对甲壳聚多糖的急性毒性以及长期毒性进行研究。方法:按照中药新药临床前研究指导原则。结果:甲壳质口服最大耐受剂量,分别相当于临床用量的750~1500倍和300~600倍。长期毒性试验表明:大鼠外观、行为、摄食、排便均无明显改变,生理生化指标及病检结果也未见明显变化。用甲壳质80mg/kg,160mg/kg,分别相当于推荐剂量20~40倍和40~80倍,对小鼠神经系统无明显影响;40mg/kg,80mg/kg分别相当于临床剂量10~20倍和20~40倍,对大鼠心血管系统和呼吸系统均无明显影响。结论:甲壳质在超常剂量情况下,毒性(急毒、长毒)几乎没有。甲壳质是一种安全的天然药物,可以长期使用。 Objective:To observe the chronic toxicity and acute toxicity of Chintin. Methods:According to the guidebook of Chinese Traditional Medicine Pre—Clinical Trial. Results:The maximum endurable oral dosages of Chitin are 750~1500 times and 300~600 times of the clinical dosages respectively. Chronic toxicity experiments show us:the appearance behavior appetite and defecation of the rats have no evident change, the biochemical and physiological indexes and the results of pathologic checking have no evident change either. 80mg/kg and 160mg/kg which are amount to 20~40 times and 40~80 times of the clinical dosages recommended have no effect on mice neural system. 40mg/kg and 80mg/kg which are amount to 10~20 times and 20~40 times of the clinical dosages recommended have no effect on cardiovascular system and respiratory system of rats. Conclusion: Chitin doesn't have any toxicity (chronic or acute). Our research work tells us that Chitin is a safe natural drug, which can be administered for a long—term.
