摘要
目的 :探讨左旋四氢巴马汀 (L Tetrahydropalmatine,L THP)对脑复苏炎症反应的影响及其治疗作用。方法 :采用四血管阻塞的方法 ,复制出大鼠全脑缺血再灌注模型。采用免疫组化法、原位杂交法和原位末端标记法分别检测假手术组 (A组 )、全脑缺血再灌注组 (B组 )、左旋四氢巴马汀治疗组 (C组 )海马CA1区核因子κB(NF κB)、白介素 1βmRNA(IL 1βmRNA)及细胞凋亡数的变化。结果 :与A组比较 ,B组海马CA1区NF κB和IL 1βmRNA的表达及细胞凋亡数明显增加 (P <0 .0 1) ;NF κB和IL 1βmRNA的表达分别于再灌注 6和12h达到高峰 ,并持续到4 8h ;细胞凋亡数随再灌注时间的延长而逐渐增多 (P <0 .0 1)。L THP治疗后 ,NF κB和IL 1βmRNA的表达下降 ,细胞凋亡数减少 (P <0 .0 1)。结论 :在脑复苏救治过程中 ,L THP可抑制NF κB与IL 1βmRNA的表达 ,抑制炎症反应 ,减少细胞凋亡而发挥脑保护作用。
Objective: To explore the protective effects of L-Tetrahydropalmatine (L-THP) on cerebral resuscitation. Methods: The model of global cerebral ischemia-reperfusion was established by four-vessel occlusion method. Using immunohistochemical technique, in situ hybridization and in situ end labeling technique (TUNEL), the expressions of NF-κB and IL-1βmRNA and the number of the apoptotic cells were examined in the sham-operated group (group A), the control group (the group after global cerebral ischemia-reperfusion treated with saline, group B) and the group treated with L-THP (the group after global cerebral ischemia-reperfusion treated with L-THP, group C) separately. Results: Compared with group A, the expression of NF-κB and IL-1βmRNA in group B was increased significantly ( P <0.01), which peaked at 6 h and 12 h respectively. With the time of reperfusion prolonged, the number of apoptotic cells was gradually increased ( P <0.01). Compared with group B, the expression of NF-κB and IL-1βmRNA and the number of apoptotic cells in group C were decreased ( P <0.01). Conclusion: L-THP may down-regulate the expression of NF-κB and IL-1βmRNA after cerebral resuscitation. It inhibited the inflammatory reaction and attenuated cellular apoptosis.
