大鼠血浆中龙胆苦苷的检测

DETERMINATION OF GENTIOPICROSIDE IN RAT′ S PLASMA

目的 建立大鼠血浆中龙胆苦苷的检测方法。方法 采用RP—HPLC法测定大鼠血浆中药物浓度 ,色谱柱为LichrospherC18(4 .6× 2 5 0mm ,5 μm) ,流动相为甲醇 -水 (33:6 7V/V)。 结果 在 1～ 2 0mg .L- 1范围内龙胆苦苷峰面积与浓度线性关系良好 (r =0 .9997) ,最低可定量浓度为 0 .0 3mg .L- 1,绝对回收率为 88.4 %～ 96 .2 % ,准确度为 97.8%～ 10 4 .9% ,日内、日间RSD均小于 10 %。结论 本测定方法灵敏、准确、简便 ,适合于龙胆苦苷的药代动力学研究。

Aim To develop a simple,sensitive and rapid method for the determination of gentiopicroside in rat′s plasma. Method The plasma concentration of gentiopicroside was determined by RP-HPLC method ,the analysis involved a 250mm × 4.6mm column packed with C 18 (5um),the mobile phase consisted of methyl alcohol-water(33∶67) Result A good linearity was obtained in the range of 1～20 mg. l-1 (r=0.9997),the recovery was between 88.4～96.2%, the accuracy was between 97. 8%～104.9% .The relative stand deviations of within-day and between day were less than 10%,the minimal detectable concentration in rat′s plasma was 0.03 mg. l-1 Conclusion The result demonstrated that the method was sensitive,accurate and simple for the determination of gentiopicroside in rat′s plasma,it is suitable for the pharmacokinetics study of gentiopicroside.
