摘要 采用HPLC法对红曲霉发酵样品中的麦角固醇 (Ergosterol)进行测定。色谱柱为Shim_packVP -ODSC18(4.6× 15 0mm ;5 μm)柱 ,以甲醇—水 (97:3)作为流动相 ,流速1.8ml/min ,紫外检测波长 2 82nm。红曲霉发酵样品经碱性乙醇皂化、乙醚萃取后 ,用外标法进行定量检测。结果表明 :Ergosterol标准曲线在 0 .0 2mg/ml~ 1.6mg/ml范围内呈线性关系 ,r =0 .994 9,该法的回收率为 98.86 % ,相对标准误差为 0 .15 %。本测定方法操作简便、准确、可靠。 The detection of Ergosterol in the fermentation sample of Monascus by HPLC was studied. A Shim_pack VP_ODS C18(4.6×150mm;5μm) column was used. The mobile phase was as follows: methanol: water=97:3(v/v) and the flow rate was 1.8mL/min. It was measured by an ultraviolet detector under 282nm.. The fermentation sample of Monascus was saponified by alkaline ethanol and then extracted by aether.. The results show the calibration curve of Ergosterol was linear with range of 0.02mg/ml ~1.6mg/ml, the correlative coefficient was 0.9949. The recoveries of Ergosterol were 98.86%. The average relative deviation (RSD) was 0.15%. So, it was a simple, sensitive and accurate method.
