藻酸钙复合材料构建组织工程化骨软骨的实验研究 被引量:6

摘要 目的 :对经体外培养的骨髓基质干细胞与藻酸钙及生长因子复合形成的组织工程性骨软骨进行形态学观察。方法 :以可注射性藻酸钙凝胶作为细胞载体 ,将体外扩增的BMSCs和生长因子IGF Ⅰ、TGF β分为 5种组合 ,注射于自体新西兰白兔皮下 ,分期取材观察。结果 :藻酸钙凝胶复合BMSCs及IGF Ⅰ和TGF β能在皮下形成软骨组织 ,逐渐转化为骨组织并最终被吸收 ,无免疫排斥及毒性反应。结论 :BMSCs 藻酸钙 生长因子复合材料能形成软骨及骨组织。藻酸钙适合BMSCs生长 ,是一种良好的软骨组织工程载体。 Objective:To observe histomorphology of neo-osteochondral tissue obtained with tissue engineering by combining calcium alginate gel and bone marrow stromal cells and growth factors.Method:Forming five groups by combining calcium alginate,stem cells and growth factors,we injected five group compound materials into the dorsal subcutaneous tissue in rabbit.Specimens were harvested at 4 th ,8 th ,12 th week after injection.Result:Bone marrow stem cells were capable of differentiation to chondroblasts or osteoblasts in the calcium alginate gel and finally regenerate cartilage-like and bone combined with IGF-Ⅰ and/or TGF-β without immunological reject and toxic reaction.Conclusion:Calcium alginate+stem cells+growth factors composite may succeed in constructing engineered osteochondral tissue.Alginate could serve as a carrier to fill osteochondral defects in vivo.The biodegradable alginate appears to provide the satisfactory 3-dimensional support in where is conducive to osteochondral tissue.
