川芎嗪对骨髓移植后小鼠LFA-1和ICAM-1表达的影响 被引量:5

摘要 本研究探讨川芎嗪对骨髓移植 (BMT)后小鼠骨髓中LFA 1和ICAM 1表达水平的影响及其促进骨髓造血重建的机制。 15 0只BALB/c小鼠随机分为正常组、生理盐水组和川芎嗪组。正常组未作任何处理 ,生理盐水组和川芎嗪组在BMT后分别喂饲相同剂量的生理盐水 ( 0 .2毫升 /只 ,每天 2次 )和川芎嗪 ( 2毫克 /只 ,每天 2次 ) ,并分别于BMT后第 7,14 ,2 1,2 8天统计存活率 ,计数脾集落形成单位 (CFU S)、外周血细胞、骨髓单个核细胞 (BMM NC) ,分析骨髓组织学变化及LFA 1和ICAM 1表达水平。结果表明 :川芎嗪组小鼠在BMT后第 10天CFU S计数和BMT后第 7,14 ,2 1,2 8天存活率、外周血白细胞、血小板、BMMNC计数、骨髓造血组织容量以及LFA 1表达水平均显著高于生理盐水组 (P <0 .0 1或P <0 .0 5 ) ,成熟红细胞容量和ICAM 1表达水平均明显低于生理盐水组 (P <0 .0 1或P <0 .0 5 )。川芎嗪组小鼠脂肪组织容量在BMT后第 7、14天明显高于生理盐水组 (P <0 .0 1) ,在第 2 1,2 8天明显低于生理盐水组 (P <0 .0 1)。结论 :川芎嗪改善骨髓微环境 ,促进造血重建。 The objective of study was to investigate the effect of ligustrazine on the expression of LFA 1 and ICAM 1 in bone marrow and the mechanism of hematopoietic reconstitution after bone marrow transplantation (BMT). The 150 mice were randomly divided into 3 groups: normal group, saline group and ligustrazine group. The normal group was not treated. The saline group was given normal saline (0.2 ml/mouse, twice a day) through gastric tube, while the ligustrazine group was given ligustrazine (0.2 ml/mouse, twice a day) through gastric tube. At days 7,14,21 and 28 after BMT, the survival rate, colony forming unit of spleen (CFU S), peripheral blood cells, bone marrow mononuclear cells (BMMNC), histologic changes of bone marrow, as well as the expression level of LFA 1 and ICAM 1 were observed. The results showed that in ligustrazine group CFU S counts on day 10 and survival rate, WBC and Plt amount in peripheral blood, BMMNC counts, hematopoietic tissue volume as well as the expression level of LFA 1 at 7,14,21,28 days after BMT were higher than in saline group (P<0.01 or P<0.05). However, mature RBC volume and expression level of ICAM 1 were lower than in the saline group (P<0.01 or P<0.05). Furthermore, fat tissue volume was higher at 7 and 14 days (P<0.01) and was lower at 21 and 28 days (P<0 01) after BMT than in the saline group. It is concluded that ligustrazine could improve bone marrow microenvironment and promote hematopoietic reconstitution.
