摘要
目的 探讨银杏叶提取物 (EGb)对急性重症胰腺炎 (ASP)胰腺细胞凋亡的作用机制。方法 将 78只SD大鼠分为假手术 (SO)组、ASP组和治疗组。治疗组在ASP模型诱发后经腹腔注射金纳多注射液 2 0mg/kg ,每 6h注射一次 ,动态观察胰腺病理变化 ,免疫组化 (TUNEL法与SP法 )检测胰腺凋亡情况与细胞凋亡调控基因Bcl 2和C myc蛋白的表达。结果 治疗组胰腺病理损害程度较ASP组减轻。ASP组凋亡指数明显高于SO组 ,(P <0 0 1) ,且术后 3h达高峰 ,治疗组 3h、6h凋亡指数明显下降 (P <0 0 5 )。免疫组化 :ASP组Bcl 2表达明显增多 ,治疗组 3h的Bcl 2表达较ASP组同时相点上调 (P <0 0 5 ) ;ASP组C myc表达术后 6h达高峰 ,治疗组 3h、12h的C myc表达虽有增高 ,但无显著性差异。结论 金纳多能减轻胰腺病理损害、抑制胰腺细胞的凋亡 ,其抑制细胞凋亡可能与其上调凋亡调控基因Bcl 2的表达有关。
Objective To investigate the effects of ginkgo biloba extract on apoptosis of pancreatic acinar cells in acute severe pancreatitis in rats.Methods Seventy-eight SD rats were randomly divided into three groups:sham operated group,ASP group and EGb treatment group.Treatment group received peritoneal injection of ginaton injection solution in a dose of 20mg·kg -1 immediately after the model of ASP being induced,then it was given injection of the same dose per 6 hours.The pathological alterations in pancreatic tissues were also examined with microscope and electronic microscope.Apoptosis of pancreatic acinar cells was detected by TUNEL method.The expression of apoptosis-regulated gene Bcl-2 and C-myc were detected by immunohistochemical technique.Results The pathological damage of pancreatic tissues were obviously alleviated in animals treated with ginaton.Results from the TUNEL staining showed that the apoptotic index of pancreatic acinal cells in treated group was obviously lower than that of ASP group( P< 0.05).Immunohistochemical analysis showed that the positive rate of Bcl-2 protein in ginaton group at 3 hours after treatment was significantly higher than that of ASP group( P< 0.05),but there was no significant difference on the positive rate of C-myc protein between treated group and ASP group.Conclusions Our data suggest that ginaton can reduce the damage of pancreatic tissues in ASP,and can restrain the speed of apoptosis of pancreatic acinar cells,this effect might be related with the expression of Bcl-2.
