摘要
对平托花生叶片、茎段、带腋芽茎段离体组织培养的研究表明,茎段培养21 d后逐渐褐化、枯死;带腋芽茎段在MS+0.5 mg·L^(-1)NAA+3.0mg·L^(-1)6-BA培养基上可直接诱导出丛生芽;叶片在MS+0.5mg·L^(-1)NAA+3.0mg·L^(-1)6-BA培养基上可成功诱导出愈伤组织,进而再生出众多的芽并进一步伸长。分离的正常幼芽在MS+0.5mg·L^(-1)NAA培养基上培养15d后长出不定根,并长成正常的平托花生植株。
The leaf .stem and stem segment with axillary bud of A. pintoi were used for the study on tissue culture. The results showed that the stem segment turned brown and wilted gradually after twenty-one days;but for stem with axillary bud,the clumped buds could be directly induced after inoculated on MS + 0. 5 mg·L-1 NAA + 3. 0 mg· L-1 6-BA medium. The leaf explants inoculated on MS + 0..5 mg· L-1 NAA + 3. 0 mg· L-1 6-BA medium, and were induced embryogenic callus, then formed many clumped buds and then differentiated successfully the plantlets; the excised plantlets could produce root after inoculated on MS + 0. 5 mg· L-1 NAA medium after fifteen days and developed into normal plants gradually.
