槲皮素诱导Cloudman S91细胞凋亡的机制 被引量:10

The potential mechanism of quercetin on apoptosis induction in mice melanoma Cloundman S91 cells

摘要 目的 :探讨槲皮素诱导小鼠黑色素瘤CloudmanS91细胞系凋亡的作用和机制。 方法 :采用噻唑蓝(MTT)比色法、吖啶橙 /溴乙锭荧光染色、流式细胞仪及westernblot分析技术对槲皮素诱导CloudmanS91细胞凋亡的作用及机制进行分析。 结果 :槲皮素对CloudmanS91细胞增殖有明显的抑制作用 ,其细胞存活率呈时间 剂量依赖性降低 ;用 4 0~ 1 2 0 μmol/L的槲皮素作用CloudmanS 91细胞 72h ,可见明显的细胞凋亡形态学变化 ;流式细胞仪检测有亚G1 峰出现 ,细胞被阻滞于G0 /G1 期 ;westernblot分析提示槲皮素可抑制 p5 3、bcl 2基因的表达 ,且呈显著的剂量依赖性。 结论 :槲皮素诱导CloudmanS91细胞的凋亡 ,其机制可能是通过阻滞细胞于G0 /G1 期、抑制 p5 3、bcl Objective:To explore the biological effect of quercetin on mice melanoma Cloundman S91 cells. Methods: MTT reduction assay, fluorescence straining with acridine orange (AO)/ethidiumbromide (EB) morphologically , flow cytometry analysis technique and Western blotting technique were performed. Results:The inhibition of quercetin on the growth of Cloudman S91 cell line was observed in a concentration and time dependent manner. The cells treated with 40~120 μmol/L quercetin for 72 hours showed a typical apoptotic morphology and hypodiploid peak before G 1 phase, Quercetin arrested Cloudman S91 cells in G 0 /G 1 phase. Quercetin strongly inhibited the expression of by p53,bcl 2 gene proteins in a dose dependent manner after 48 hours treatment. Conclusion:quercetin induced of mice melanoma Cloudman S91 cells apoptosis, which may be related to the arrest G 0 /G 1 phase and inhibition of p53,bcl 2 gene proteins in Cloudman S91 cells.
