摘要
目的 研究抗氧化剂EseroS -GS对巨噬细胞iNOS基因表达的抑制作用 ,并探讨其作用的机制。 方法 本项研究以脂多糖和γ -干扰素诱导巨噬细胞表达可诱导型一氧化氮合酶 ,应用蛋白质印迹分析以及ESR自旋捕集技术研究了EseroS -GS对巨噬细胞产生内源性一氧化氮的作用。 结果 研究结果表明EseroS -GS能够抑制巨噬细胞可诱导型一氧化氮合酶基因表达、减少内源性一氧化氮的生成 ,而这一抑制作用是通过NF -κB信号通路实现的。 结论 EseroS -GS对巨噬细胞的活化具有显著的调控作用。
Objective The regulation of antioxidant EseroS-GS on the expression of inducible nitric oxide synthase in macrophages was studied for revealing the mechanism of EseroS-GS. Methods (1) The expression of inducible nitric oxid synthase in macrophages by using LPS/IFN; (2) The effects of EseroS-GS on the generation of superoxide anion and nitric oxide in stimulated primary macrophages was studied by Western blot analysis and ESR techniques. Results The result shows that EseroS-GS scavenges the endogenous superoxide anion dose-dependently, inhibited the expression of iNOS gene via modulating the NF- _κB signaling pathway, and decreases the formation of nitric oxide. Conclusions Our data demonstrates that EseroS-GS plays a modulation role in the activation of macrophages.
