黄芩素甙对沙土鼠脑缺血海马微管运动蛋白Kinesin活性的影响

Effect of breviscapine(黄芩素甙) on the activity of Kinesin in hippocampus in gerbils during cerebral ischemia/reperfusion

摘要 目的 :观察黄芩素甙对脑缺血再灌注期间海马锥体细胞微管运动蛋白 Kinesin活性变化的影响。方法 :制备沙土鼠前脑缺血再灌注模型 ,脑缺血时间为 10 m in。应用免疫组织化学染色方法结合计算机图像分析技术 ,测定海马微管运动蛋白 Kinesin的活性。结果 :在海马 CA 1区 ,常温对照组缺血再灌注 6、4 8和96 h时微管运动蛋白 Kinesin活性分别降至假手术组的 5 8%、38%和 12 % (P均 <0 .0 1) ,而黄芩素甙组在再灌注 6、4 8和 96 h后 ,微管运动蛋白 Kinesin活性分别为假手术组的 81%、6 1%和 2 1% ,均明显高于常温对照组(P均 <0 .0 5 )。在海马 CA2、CA3和 CA4区 ,微管运动蛋白 Kinesin的活性无明显变化。结论 :黄芩素甙可通过抑制脑缺血再灌注期间海马 CA1区微管运动蛋白 Kinesin活性的下降来达到脑保护作用。 Objective: To investigate the effect of breviscapine on the activity of Kinesin (a microtubule based motor protains) in the hippocampus in gerbils during cerebral ischemia/reperfusion. Methods: The cerebral ischemia/reperfusion models of gerbils were achieved by occlusion bilateral carotid arteries. Cerebral ischemia lasted for 10 minutes. The activity of Kinesin in the hippocampus was determined at 6 hours, 48 hours, 96 hours after reperfusion by immunohistochemical staining with computer image analysis system. Results: The activities of Kinesin in the hippocampus CA1 sector in gerbils in normathermia groups significantly decreased after reperfusion, and its activities at 6 hours, 48 hours, 96 hours reperfusion were about 58%, 38%, 12% of that in shamoperated groups(all P<0.01). However, the activities of Kinesin in breviscapine groups were about 81%, 61%, 21% of that in sham-operated groups. Breviscapine obviously mitigated the decrement of the Kinesin activity in the hippocampus CA1 sector in gerbils(all P<0.05). No marked changes in Kinesin activity in the hippocampus CA2, CA3, CA4 sector were observed during cerebral ischemia/reperfusion. Conclusion: Breviscapine can protect cerebral function through significantly inhibiting the decrease in Kinesin activity after cerebral ischemia/reperfusion in gerbils.
