摘要
目的:研究麦全冬定对被动吸烟大鼠脑组织诱导型一氧化氮合酶(induciblenitricoxidesynthase,iNOS)表达的影响。探讨麦全冬定在吸烟导致的脑缺血损伤中的保护作用机制。方法:采用免疫组织化学ABC方法检测脑组织iNOS活性。结果:①正常对照组:大鼠大脑皮质、海马、纹状体均有少量iNOS表达(7.89±0.40),(10.16±2.99),(7.04±0.52)个/视野。②吸烟组:各脑区iNOS表达明显升高(37.16±2.82),(43.07±1.24),(39.50±3.33)个/视野(t=22.22,13.95,11.92,P<0.01)。③麦全冬定组与吸烟组比各脑区iNOS表达显著降低(20.90±4.02),(28.95±1.61),(15.33±2.36)个/视野(t=3.22,2.82,11.31,P<0.05)。结论:吸烟通过脑组织中iNOS表达增强生成过多的一氧化氮,造成脑神经细胞毒性损伤,麦全冬定对iNOS表达有下调作用,从而减少脑神经细胞毒性损伤。
AIM:To study the effects of vedrin on the activity of inducible nitric oxide synthase (iNOS) in the brain of passive smoking rats. METHODS:ABC immunohistochemical method was applied to measure the activity of iNOS in the brain. RESULTS:There were a few expressions of iNOS in cerebral cortex[(7.89±0.40) per visual field],hippocampus[(10.16±2.99) per visual field]and corpus striatum[(7.04±0.52) per visual field] in the control group.Compared with the control group, the expression of iNOS in the smoking group was significantly increased[(37.16±2.82),(43.07±1.24),(39.50±3.33) per visual field respectivley(t=22.22,13.95,11.92,P< 0.01].In addition, the expressions of iNOS in the vedrin group[(20.90±4.02),(28.95±1.61),(15.33±2.36) per visual field respectively]were significantly lower than those in the smoking group(t=3.22,2.82,11.31,P< 0.05). CONCLUSION:Cigarette smoking can cause cytotoxic lesion owing to the excessive produce of nitric oxygen through the increased expression of iNOS in the brain.Vedrin can reduce the expression of iNOS so as to alleviate the cytotoxic lesion.
