氧化苦参碱对大鼠肝星状细胞旁分泌活化途径的抑制作用 被引量:27

摘要 目的 观察氧化苦参碱对大鼠激活肝星状细胞 (HSC)这一旁分泌活化途径的影响。方法 分离正常及CCl4损伤肝库普弗细胞 (KC) ,进行体外培养 ,收集损伤肝KC条件培养液 (IKCM )及药物 (氧化苦参碱 ,Oxy)干预后的条件培养液 (Oxy IKCM ) ,以IKCM及Oxy IKCM添加于原代培养的未活化HSC培养体系中 ,观察Oxy对KC旁分泌激活HSC的影响 ,生物学方法检测Oxy对活化的KC分泌TGFβ1的影响。结果 急性损伤肝KC对HSC的增殖有促进作用 ,氧化苦参碱可抑制KC对HSC增殖的促进作用 ,并可抑制活化KC分泌的TGFβ1。结论 氧化苦参碱可通过抑制旁分泌途径抑制HSC活化及活化KC分泌的TGFβ1。 Objective To investigate the effect of oxymatrine(Oxy) on the activation of hepatic stellate cells(HSC) through paracrine stimulation by Kupffer cells(KC). Methods Hepatic Kupffer cells and hepatic stellate cells from rats were isolated and cultured. Conditioned culture medium of Kupffer cells(KCM) and Oxy treated KCM(Oxy-KCM) from acute injured rat liver induced by CCl 4 were collected. KCM and Oxy-KCM were added to the primary culture quiescent HSC medium, after co-cultured for 48 and 72 hours, the proliferation of HSC were investigated by MTT colorimeter assay. Secretion of TGFβ1 by KC was assayed by biological test. Results Oxymatrine can inhibited the proliferation of HSC through paracrine stimulation of activated Kupffer cells by suppressing TGFβ1 secrtion. Conclusion Oxymatrine can inhibit the proliferation of HSC through paracrine stimulation by suppressing the TGFβ1 secreted by activated Kupffer cells.
