摘要
本文介绍一种非酶体系用核黄素,四甲基乙二胺光照产生O(?)氧化羟胺形成亚硝酸盐的原理(?)建立测 SOD 活性的新方法,并对测活体系的组分浓度、吸收波长、光照时间等进行了务件测试。本方法具有快速、简便、灵敏、稳定、价廉、受干扰小,不需特殊仪器设备,便于实际运用等优点。
Superoxide dismutases(SOD ECL 15.11)are a broad class of metalloenzymes,character-ized by the property of undergoing the dismuta-tion of superoxide anion free radical(O^-_2).Thereare three kinds of such enzymes,such asCu,Zn-SOD,Fe-SOD and Mn-SOD.Numerousmethods of assay of SODs have been reported sofar.We introduce a novel method using a non-enzyme system containing riboflavin andTEMED.The superoxide anion free radical(O_2)can be generated by illumination,and ni-trite formation can be followed when hydroxy-lamine exists in the system.The optimal condi-tions of the system,such as composition concen-tration,absorbance wavelength,and time forphotosensization are also studied.Satisfactoryresults can be obtained when the system is ap-plied to the assay of Cu,Zn-SOD and Mn-SODis isolated from microgonisms by the assay of theGeluomeng(a healheare food).The method issimple,fast,sensitive,and free from seriousinterferences.
