摘要
本文用 HPLC 法直接测定尿中呋喃妥因浓度。最低检出浓度为0.1μg/ml,日内、日间差异<4.6%。体外实验表明,氟哌酸、TMP、SMZ 均不干扰测定结果。尿样在冷冻(-10℃)条件下2(?)保持稳定。
An analytical method was established for determining urine concentration of nitrofuran-toin(NFT)using HPLC.The mobile phase consisted of 20/80 by vol methanol/0.01 mol/LNa acetate,using glacial acetic acid adjusting thepH of the solution to 5.0,and then adjusting pHto 6.69 with ethylenediamine.The wavelengthof detection was fixed at 254nm.20μl of 4 mg/ml internal standard solution(furazolidone)wasadded to 1 ml of the urine sample,that wasshaked well and injected directly into the chro-matograph.The limit of detection for NFT inurine was 0.1 μg/ml.The standard curve ofNFT was linear within the concentration range 2~200 μg/ml.The within-day and between-dayvariation coefficienes were less than 4.6%.Norfloxacin,TMP and SMZ did not interferewith determination of NFT.Urine samples keptfrozen at — 10℃ were stable during the 20days.
