摘要
在空腹和食物条件下分别给7名健康志愿者交叉口服呋喃妥因(nitrofurantoin;NFT)肠溶片,从尿药排泄量估算生物利用度。结果表明:食物可明显降低该制剂的生物利用度。24h 尿药总排泄量:空腹为15.4%±5.43%,食物条件为1.85%±0.68%(P<0.001);空腹条件尿中最大药物浓度为107±55μg/ml,而食物条件仅为13.9±7.8μg/ml,低于最低杀菌浓度(32μg/ml)。
The enteric-coated tablets of nitrofurantoin100mg were given to 7 healthy volunteers underthe conditions of fas.ting and after meal.The urine concentration of ni-trofurantoin was determined by method of HPLC.The results show that food can significantly reducethe bioavailability of nitrofurantoin.The totalamount of nitrofurantoin which excreted within 24hwas 15.4%±5.43% in fasting condition and1.85%±0.68% after meal(P<0.001),themaximum concentration of nitrofurantoin in theurine was 13.9±7.8μg/ml after meal,which isbelow the lowest limit.of bactericidal concentration(32μg/ml).We conclude that enteric-coated tabletsof nitrofurantoin should be administered on fastingcondition.
