HPLC测定维胺酯在大鼠体内的血药浓度及其药代动力学

Plasma HPLC determination and pharmacokinetics of Vitaminate after Ⅳ injection in rats

本文用高效液相色谱法测定了大鼠体内维胺酯[N-(4-乙氧羰基苯基)-维甲酰胺]的血药浓度,用乙腈直接稀释血浆沉淀蛋白,用 Zorbax-C_ 8柱作固定相,以90%甲醇水溶液作流动相,370nm 波长为检测波长,用外标法峰面积定量,在0.2～15μg/ml 的浓度范围之间峰面积与浓度呈线性(r=0.9997),最低检测浓度为0.1μg/ml。大鼠静脉注射维胺酯后测得药代动力学参数α、β、Vc、C(?)分别为3.83h^(-1)、0.19h^(-1)、0.22L/kg、0.27L·h^(-1)·kg^(-1)。

Vitaminate[N-(4-ethoxycarbophenyl)reti-namide]is a kind of new retinamide.A HPLCmethod was developed for the quantitative determi-nation of vitaminate in rats plasma.The chromato-graphic conditions included a zorbax—C_8(5μm)re-versed-phase column(4.615);methanol-water(90:10)mobile phase;UV detector at 370nm.Goodlinear range was 0.2～15μg/ml(r=0.997).Thelowest detective concentration was 0.1μg/ml.Afterintravenous injection of vitaminate to rat,the plas-ma lever decreased in two phases:distribution phaseand elimination phase with T(1/2) of 0.18h and3.68h respectively.The apparent volume of distri-bution was 1.42L/kg.
