螺旋藻多糖的纯化和高效液相色谱分析 被引量:9

摘要 采用两种改进除蛋白方法得到的螺旋藻粗多糖 ESPS和 TSPS,分别经 DE- 5 2柱层析和 Sephadex G-10 0柱层析分离纯化得精制多糖 ESPS1、ESPS2、ESPS3和 TSPS1、TSPS2 ;用高效凝胶渗透色谱法鉴定了各精制多糖的纯度并测定了分子量 ;各精制多糖经酸完全水解后 ,用高效液相色谱法测定了多糖的单糖组成 ,证明均为不同己糖构成的杂多糖 ,并且首次在螺旋藻多糖的单糖组成中发现有 Two kinds of crude polysaccharides (EPS AND TSPS) were extracted from the powder of spirulina platensis, deproteinized by two different methods (enzymolysis+sevage's method and trichloroacetic acide's method)..ESPS and TSPS were further purified by loading on DE-52 and Sephadex-100, three polysaccharide fractions of ESPS (ESPS1, ESPS2, ESPS3) and two fractions of TSPS(TSPS1,TSPS2) were obtained.Their homogeneity was examined by high performancegel permeance chromatography (HPGPC).Their molecular weights were determined by HPGPC.The composition identified by HPLC showed that the five fractions were all composed of different hexoses.D-fructose was first found in the SPS.
