摘要
目的 研究颅内血肿后脑组织线粒体功能及结构的变化。方法 以 5 0 μl鼠尾血注入大鼠尾状核制作大鼠颅内血肿模型 ,注血后分别于 12h、1、3、7d处死大鼠 ,断头取血肿周围脑组织 ,测定以上不同时相点血肿周围组织线粒体膜流动性 ,并在电镜下观察线粒体结构变化。结果 颅内血肿早期 ( 12h)线粒体膜流动性并无明显减退。出血后 1d线粒体膜流动性较对照组下降 (P <0 0 5 ) ;电镜下观察线粒体结构有一定变化。出血 3d及 7d后线粒体膜流动性较对照组明显下降 (P <0 0 1) ;线粒体结构明显紊乱。结论 颅内血肿早期血肿周围组织线粒体功能无明显下降 ,结构无显著变化 。
Objective To study the changes of the mitochondrial membrane fluidity of brain and its ultrastructural changes after intracerebral hematoma (ICH) in rats. Methods Rat ICH model was established by means of injection of 50 μl tail blood into the caudoputamen. Rats were sacrificed at 12 h, 1 d, 3 d, and 1 w after injection of blood. The brain tissue around the hematoma was collected for the determination of the mitochondrial membrane fluidity of rat brain around the hematoma at different time points, i.e. at 12 h, 1 d, 3 d, and 1 w after injection of blood. The ultrastructural changes of mitochondria were observed electron microscopically. Results At 12 h after injection, the membrane fluidity of cerebral mitochondria remained at a higher level ( P >0.05), but it began to decrease ( P <0.05) at 1 d after injection of blood, and decreased markedly at 3 d and 7 d after injection of blood ( P <0.01). The mitochondrial ultrastructure around the hematoma changed mildly in 1 d group, but it was severely damaged in 7 d group. Conclusion The cerebral mitochondrial function can be destroyed markedly at the late stage after ICH in rats.
