摘要
目的 探讨体外培养胎鼠下丘脑神经元的形态、生长变化规律及促肾上腺皮质激素释放激素(CRH)的表达变化。方法 取孕 1 7天胎鼠下丘脑分散培养下丘脑神经元 ,光镜下连续观察神经元胞体直径、突起长度、数目等变化规律 ,应用扫描电镜、免疫细胞化学染色方法观察CRH神经元的生长变化规律。结果 培养下丘脑CRH神经元 3~ 6天生长速度最快 ;7~ 1 0天神经元细胞数量、胞体直径、突起长度和CRH染色强度均达高峰 ;1 2天后部分神经元可出现退形性改变。结论 培养下丘脑CRH神经元 7~ 1 0天生长迅速趋于成熟 。
Objective To investigate the morphology and development of cultured fetal rat hypothalamic CRH neurons.Methods Perfused primary cultures of fetal rat hypothalamic neurons were studied by immunocytochemical technique and scanning electronic microscope.Results Under the microscope,the neurons were found to grow fast during 3 6 days,and reached the highest levels of their number,cell body size,staining intensity of CRH,and synapse length during 7 10 days. After 12 days, the number and staining intensity of CRH neurons decreased.Conclusion The hypothalamic CRH neurons maturae during 7 10 days with the best function.
