哌仑西平对鸡形觉剥夺性近视的作用 被引量:4

摘要 目的 观察选择性M1受体拮抗剂哌仑西平对鸡形觉剥夺性近视的疗效 ,初步探讨其作用机制。方法  1d龄鸡 40只 ,随机分为 :正常组 (Ⅰ ) ;单纯遮盖组 (Ⅱ ) ;药物对照组 (Ⅲ ) ;哌仑西平组 (Ⅳ )。Ⅱ、Ⅲ、Ⅳ组鸡的右眼用半透明眼罩遮盖 ,Ⅲ、Ⅳ组鸡的剥夺眼分别每日结膜下注射 0 1mol/L磷酸缓冲液和哌仑西平溶液 ;2周后检测 4组鸡的右眼屈光度、眼轴长度、赤道部直径 ,并对后极部巩膜行光镜、电镜观察。结果 哌仑西平组的各项指标较单纯遮盖组、药物对照组显著减低 (P <0 0 5 ) ,较正常组形成相对近视 ,眼轴长度、赤道部直径明显增大 (P <0 0 5 ) ;药物对照组与单纯遮盖组相比各项指标无显著性差异 (P >0 0 5 ) ;组织学观察结果表明 ,哌仑西平组后极部巩膜的病理改变较单纯遮盖组明显减轻。结论 选择性M1受体拮抗剂哌仑西平能部分抑制近视的发生发展 ,可能是通过作用于巩膜发挥疗效的。 ObjectiveTo observe the effect of M 1-selective muscarinic antagonist,pirenzepine,on form deprivation myopia.MethodsFourty one-day-old chickens were divided into 4 groups randomly:Ⅰ normal control group,Ⅱ form deprivation group,Ⅲ vehicle control group and Ⅳ pirenzepine injection group.Form deprivation myopia was established in right eyes of group Ⅱ,Ⅲ and Ⅳ by placement of a translucent occluder.The deprived eyes of group Ⅲ and Ⅳ received daily subconjunctival administration of vehicle control and pirenzepine,respectively.Optical measures such as refraction,axial length,equatorial diameter and structure observation under light and electronic microscopy were made at the end of the experiment.ResultsValues of refraction status,axial length and equatorial diameter of the eyes in pirenzepine injection group were significantly lower in comparison with the form deprivation group (P<0.05),but were higher and form relatively myopia when compared with normal control group (P<0.05) .There were no significant differences in optical measures between drug control and pirenzepine injection group (P>0.05).Histopathologic examinations indicated that pirenzepine obviously alleviated the pathologic changes of sclera.ConclusionSubconjunctival administration of pirenzepine partly prevents form deprivation-induced myopia by inhibiting scleral pathologic change.
