摘要
目的 探讨抗氧化剂维生素C对氧化损伤所诱发的的晶状体上皮细胞凋亡的抑制作用。方法 分别以不同时限观察对照组、H2 O2 氧化损伤组、氧化损伤同时加VitC共 3组的晶状体混浊情况 ,并用TUNEL法及DNA片段分析检测各组各时晶状体上皮细胞凋亡情况。结果 晶状体混浊情况显示随培养时间延长VitC组晶状体混浊度明显轻于氧化损伤组 (P <0 0 1) ;TUNEL检测发现 ,VitC组各时限凋亡率均明显低于H2 O2 组 (P <0 0 1) ;DNA片段分析显示H2 O2 组2 4、3 6、48及 72h各时限均呈现凋亡细胞典型梯状条带 ,而培养 2 4h的对照组和VitC组均未出现此变化而仅呈现正常电泳条带。结论 抗氧化剂VitC可抑制H2 O2 氧化损伤所诱导的兔晶状体上皮细胞凋亡并减轻或延缓白内障形成。
ObjectiveTo investigate the inhibitory effect of antioxidants(Vit C) on H 2O 2-induced lens epithelial apoptosis.MethodsLenses of in vitro rabbits were incubated in MEM medium,and they were divided into control group,H 2O 2 group and H 2O 2 with Vit C group.The opacity of the lenses in every group was evaluated.The apoptosis of lens epithelial cells was detected using terminal deoxynucleotidy transferase mediated x-dutpnick end labeling (TUNEL) method and DNA fragmentation assay.ResultsThe opacity and apoptosis rates in Vit C group were apparently lower than those of the H 2O 2 group.The DNA “ladder” was found in H 2O 2 group during 24-72 hours,but no DNA “ladder” in normal group and H 2O 2 with Vit C group.ConclusionThe combination of H 2O 2with Vit C can protect lens from the oxidative injury and restrain the apoptosis rate of lens epithelial cells and further,cataract formation.
