摘要
目的 :探讨白细胞介素 2 (IL 2 )对体外培养的正常孕早期滋养层细胞的增殖活性和绒毛膜促性腺激素(HCG)分泌的调节作用。 方法 :选取培养至第 4代生长良好的滋养层细胞 ,加入不同浓度的IL 2继续培养 4 8h后 ,用四甲基偶氮唑盐 (MTT)比色法测定细胞的增殖活性 ,用放射免疫法测定HCG浓度。 结果 :①与对照组相比较 ,IL 2在低浓度 (10 0U/ml、4 0 0U/ml)时促进滋养层细胞的增殖活性和HCG分泌 (P <0 .0 5 ) ;在高浓度 (16 0 0U/ml、6 4 0 0U/ml、2 5 6 0 0U/ml)时抑制滋养层细胞的增殖活性 (P <0 .0 5 ) ,而对HCG分泌无影响。②IL 2与滋养层细胞增殖性之间呈负相关 ,与HCG分泌水平之间无相关性 ;滋养层细胞的增殖活性与HCG分泌水平之间呈正相关。 结论 :①不同浓度的IL 2对滋养层细胞的增殖活性产生不同的作用 ,具有明显的量效关系 ,为病理性妊娠发病机制的研究提供了新的依据 ,也为临床抗早孕、异位妊娠的治疗提供新的思路。②IL 2可能不直接对滋养层细胞HCG的合成与释放产生调节作用。
Objective:To investigate the effects of IL-2 on the proliferation activity and HCG secretion of first trimester human trophoblastic cells in vitro. Methods:Growing -well 4th generation trophoblastic cells were selected,cultured for 48 h with IL-2 of different concentration. The MTT colorimetric assay was used to analyze the proliferation activity, and radioimmunoassay was used to analyze the secretion of HCG. Results:①Low concentration (100 U/ml,400 U/ml) IL-2 promoted proliferation activity and HCG secretion (P<0.05). IL-2 restrained the proliferation activity (P<0.05) and had no effect on HCG secretion in high concentration(1 600 U/ml,6 400 U/ml,25 600 U/ml). ②There was a negative correlation between IL-2 and the proliferation activity. There was no correlation between IL-2 and HCG secretion. There was a positive correlation between the proliferation activity and HCG secretion. Conclusion:①The effects of IL-2 on the proliferation activity and HCG secretion of trophoblastic cells were differed with different concentration, the dose-effect relationship was obvious. It provides new evidence for the mechanism of pathologic pregnancy and new way for therapy of early pregnancy and ectopic pregnancy.② IL-2 may not adjust the synthesis and the release of HCG of trophoblstic cells directly.
