给大鼠第三脑室微量注射δ-阿片受体激动剂DPDPE(1μg或3μg/3μl),用放射免疫法测定血浆ACTH.β-内啡肽和催乳素的含量。观察到DPDPE可显著升高大鼠血浆中上述激素水平。实验结果表明,激活δ-阿片受体可同时兴奋上述垂体前叶激素的释放;提示应激或某种病理状态下,ACTH、β-内啡肽和催乳素的相伴释放机制可能与δ-阿片受体的激活有关。
To evaluate a possible physiological and pathological role of brain δ-opiate receptor and endogenous ligands in the control of ACTH, β-endorphin (β-EP) and prolactin (PRL) release, a specific agonist of the δ-opiate receptor—[D-Pen^2, D-Pen^5]-enkephalin (DPDPE) was microinjected into the third cerebral ventricle (1 μg or 3 μg/3 μl/rat)in conscious freely-moving male rats. Plasma ACTH, β-EP and PRL concentrations were measured by radioimmunoassay (RIA). Our results clearly indicate that third ventricular injection of DPDPE significantly elevates plasma ACTH,β-EP and PRL levels in comparison with control values, and that the stimulatory effects of DPDPE on the release of the three hormones are dose-related at 1 and 3 μg. The data suggest that activation of the δ-opiate receptor may be a mechanism behind the concomitant release of ACTH, β-EP and PRL during stress and certain pathologic conditions.
