摘要 目的 利用基因表达谱芯片研究T淋巴细胞株 (HUT78)在褪黑素作用后免疫相关基因表达的差异性。方法 用Cy5和Cy3两种不同的荧光染料通过逆转录反应将干预组和对照组的HUT78细胞的mRNA分别标记成两种探针 ,并与载有一组靶基因的基因表达谱芯片进行杂交 ,通过扫描荧光强度 ,计算机软件分析 ,寻找经褪黑素作用后表达有差异的基因。结果 T淋巴细胞株受褪黑素作用 2 4h后 ,共筛选出 42条差异表达基因 ,其中 3 5条基因上调 ,7条基因下调。结论 褪黑素激活的人淋巴细胞 。 Objective To in ve stigate the difference of immuno-related gene expressions in T lymphocyte line (HUT78) incubated with melatonin by cDNA microarray of gene expression profile. Methods Two cDNA probes labelled with Cy3 or C y5 fluorescence dyes were prepared from mRNA of melatonin-treated or untreated HUT78 cells and were hybridized with cDNA microarray of target gene expression p rofile which were scanned for fluorescent intensity. The difference of gene expr ession profile was screened by computer software. Results After T lymphocyte line was incubated with melatonin for 24 h, 42 ge nes showed different expressions, among them 35 of immuno-related genes were up regulated and 7 were downregulated. Conclusion Human lymphocytes activated by melatonin play the immuno-regulatory roles throu gh regulation of a number of gene expressions.
