摘要
目的 :比较LPSip所诱导的SLE模型小鼠血清中自身抗体的表达 ,获得死亡率较低的SLE模型 .方法 :BALB/c小鼠随机分组ipLPS或生理盐水 ,2 4h后取血清行间接免疫荧光法及可提取性核抗原 (ENA)多肽抗体免疫印迹法进行实验 .结果 :LPS 5mg·kg-1ip动物死亡率为 4 0 % ;2 .5mg·kg-1死亡率为 5 % ;1.2 5mg·kg-1无死亡 .ENA多肽抗体免疫印迹法 :LPS 5mg·kg-1ip动物可见阳性条带出现于 2 8/ 2 9,13.5ku多肽 ;15 ,16 .5 ,38ku多肽 ;5 2ku多肽及4 7/ 4 8,4 5ku多肽上 ,可判定为抗Sm ,抗Rib ,抗ssA和抗ssB抗体阳性 .与LPS 2 .5mg·kg-1ip组一致 .间接免疫荧光法 :LPS 5mg·kg-1ip与 2 .5mg·kg-1组动物血清中均能检测出核糖体 (Rib) p 蛋白抗体、Jo 1抗体和线粒体 (AMA)抗体阳性 ,荧光强度较强 .1.2 5mg·kg-1组无阳性结果 .结论 :LPS2 .5mg·kg-1ip能成功诱导SLE模型 。
AIM: To compare the expressions of serum autoantibodies of systemic lupus erythematosus (SLE) models by ip injection of lipopolysaccharides (LPS) with different concentrations and to establish an animal model for SLE with lower mortality. METHODS: BALB/c mice were randomly divided into four groups and were given three different LPS doses or saline respectively. Mice serum was isolated and examined by indirect immunofluorescence and ENA immunoblot 24 hours after the injection. RESULTS: The mortality rates with LPS i.p. injection were about 40% in 10 mg·kg -1 group, 5% in 5 mg·kg -1 and zero in 2.5 mg·kg -1 respectively. By ENA immunoblot, the positive blots of 5 mg·kg -1 and 10 mg·kg -1 groups were at 28/29,13.5 ku (anti Sm Ab), 15,16.5,38 ku (anti Rib Ab), 52 ku (anti ssA Ab) and 47/48,45 ku (anti ssB Ab). By indirect immunofluorescence, the stain reaction for Rib p protein antibody, Jo 1 antibody and AMA antibody was positive in 10 mg·kg -1 and 5 mg·kg -1 groups but negative in 2.5 mg·kg -1 group. CONCLUSION: i.p. injection of 5 mg·kg -1 LPS has a lower mortality than that of 10 mg·kg -1 in establishing SLE models and it can be used as an appropriate model for further research.
