摘要
目的 :观察早期糖尿病大鼠血清对培养视网膜M櫣ller细胞 (retinalM櫣llercells,RMCs)增殖的影响 .方法 :将链脲霉素 (streptozotocin ,STZ)诱导糖尿病大鼠病程 2wk时的血清 ,以DMEM培养基稀释 (5 0 ,10 0及 2 0 0mL·L-1)后作用于体外培养的大鼠RMCs,采用四唑盐 (tetrazolium ,MTT)还原比色法 ,测定MTT反应的吸光度 (A值 ) .结果 :大鼠血清能促进RMCs增殖 ,并呈现浓度依赖性 ,而早期糖尿病大鼠血清较对照组血清 (正常同龄大鼠的同浓度血清 )有显著的抑制增殖的作用 (P <0 .0 5 ) ,并随着血清浓度的增加 ,抑制率增加 ,5 0 ,10 0及 2 0 0mL·L-1时 ,其A值分别为 0 .2 0 8± 0 .0 2 0vs 0 .2 31± 0 .0 2 0 ,0 .2 6 7± 0 .0 13vs 0 .30 4± 0 .0 14和 0 .32 7± 0 .0 2 1vs 0 .380± 0 .0 2 4 ,其抑制率分别为 10 .0 % ,12 .2 %和 13.9% .结论 :早期糖尿病大鼠血清对RMCs的增殖有显著的抑制作用 .
AIM: To observe the effect of serum from rat with incipient diabetes on the proliferation of cultured retinal Müller cells(RMCs). METHODS: A tetrazolium(MTT)reduction method was used to evaluate the proliferation of the rat RMCs cultured in DMEM with different concentrations(50,100 and 200 mL·L -1 )of sera from rats 2 weeks after streptozotocin (STZ) induced diabetes. RESULTS: In a concentration dependent manner, the sera from rats significantly promoted the proliferation of RMCs per sc ,whereas the sera from diabetic rats showed inhibitory effects on the proliferation of RMCs compared with those from age matched control rats( P <0.05), and their A values at different concentrations (50, 100 and 200 mL·L -1 ) were 0.208±0.020 vs 0.231 ±0.020, 0.262±0.013 vs 0.304±0.014 and 0.327± 0.021 vs 0.380 ±0.024, and their inhibition rates were 10.0%, 12.2% and 13.9% respectively. CONCLUSION: Serum from rat with incipient diabetes has an inhibitory effect on the proliferation of RMCs.
