摘要
目的 :观察肿瘤坏死因子 α(TNF α)和高迁移率族蛋白 1(HMG 1)基因在内毒素血症大鼠心肌组织中的动态变化 .方法 :采用RT PCR法半定量分析内毒素作用 32h内大鼠心肌组织中TNF αmRNA及HMG 1mRNA水平 .结果 :正常大鼠心肌组织可表达一定量TNF αmRNA(0 .4 1± 0 .0 8)和HMG 1mRNA(0 .5 3± 0 .0 7) .内毒素作用后 ,两者表达均增加 ,TNF αmRNA在 8h达峰值 (0 .91± 0 .0 3,P <0 .0 1) ,HMG 1mRNA在 2 4h达峰值 (1.0 3± 0 .0 9,P <0 .0 1) .结论 :内毒素能上调心肌组织中TNF αmRNA和HMG 1mR NA的表达 。
AIM: To observe the dynamic changes of tumor necrosis factor α(TNF α) and high mobility group 1 protein (HMG 1) gene expression during endotoxemia (ETM) in rat myocardium. METHODS: The levels of TNF αmRNA and HMG 1 mRNA in rat myocardium after endotoxin stimulation (0-32 h) were quantitatively analyzed with RT PCR. RESULTS: Normal rat myocardium expressed TNF α mRNA(0.41±0.08)and HMG 1 mRNA (0.53 ±0.07). When stimulated with endotoxin,both were over expressed .The peak value of TNF α mRNA in myocardium presented at 8 hours(0.91±0.03, P <0.01)while the peak value of HMG 1 mRNA was observed at 24 hours (1.03±0.09, P <0.01) after stimulation. CONCLUSION: The expression of TNF α mRNA and HMG 1 mRNA can be up regulated by endotoxin in rat myocardium.
