摘要
目的 :观察血管紧张素Ⅱ (AngiotensinⅡ ,AngⅡ )对体外培养的牛眼小梁细胞 (bovinetrabecularmeshworkcell,TMcell)形态及胶原合成的影响 ,探讨AngⅡ在原发性开角型青光眼 (primaryopen angleglaucoma ,POAG)发病机制中的意义 .方法 :AngⅡ (10 ,1,0 .1μmol·L-1)孵育TM细胞12~ 2 4h ,采用倒置显微镜观察细胞形态及计算机图像分析系统计算细胞面积 .用化学方法检测培养液中羟脯氨酸含量间接推测胶原含量 .结果 :AngⅡ产生明显细胞收缩效应 ,呈浓度依赖性 ,但细胞间连接无改变 ;AngII组细胞面积减小与对照组有显著性差异 (P <0 .0 5 ) .培养液中的羟脯氨酸含量明显升高 ,呈浓度依赖性 ,与对照组有显著性差异 (P <0 .0 5 ) .结论 :AngⅡ引起TM细胞浓度依赖性收缩 ,同时使胶原合成增强 ,是影响房水排除 。
AIM: To observe the effects of Ang Ⅱ on the morphology and collagen synthesis of cultured bovine trabecular meshwork cells in vitro and to investigate the role of Ang Ⅱ in primary open angle glaucoma (POAG). METHODS: TM cells were incubated with different final Ang Ⅱ concentrations (10, 1, 0.1 μmol·L -1 ) for 12~24 h and the morphology of TM cells was observed by inverted microscope and the area of cells was calculated by computer image analytic system. Collagen content was determined indirectly by detecting the hydroxyproline (HYP) in the culture media with chemical methods. RESULTS: Ang Ⅱ induced obvious contraction of TM cells in a dosage dependant manner, but it had no effects on cell junction. The area of cells decreased in the group with Ang Ⅱ and it showed statistical significance compared with the control group ( t ′ values 7.39, 3.83 and 2.55 respectively, P <0.01.). The HYP in the culture media increased significantly in a dosage dependant manner and it was statistically significant compared with the control group ( t ′ values 11.98, 15.70 and 14.13 respectively, P <0.01.). CONCLUSION: Ang Ⅱ induces the contraction of TM cells in a dose dependant manner and increases the production of HYP in TM cells. Ang Ⅱ may affect the aqueous outflow and may play a role in glaucoma development.
