摘要
目的 :研究高氧液对兔脊髓缺血再灌注损伤的作用 .方法 :雄性新西兰大白兔 2 0只 ,随机分成对照组 (C组 ,n =10 )及高氧液组 (H组 ,n =10 ) .夹闭腹主动脉肾下段 2 0min ,建立兔脊髓腰尾段缺血模型 .H组于缺血前 2 0min持续以 2 0mL·kg-1·h-1恒速静脉输入高氧液 ,直到再灌注后 2 0min ;C组则采用同种方法静脉输入等容量生理盐水 .测定缺血前 2 0min、缺血后 10min及再灌注 2 0min后 ,血浆中血栓素B2 (TXB2 )、6 酮 前列腺素F1α(6 keto PGF1α)的含量 ;再灌注 4 ,8,12 ,2 4 ,4 8h分别对动物后肢运动功能评分 ;再灌注4 8h后 ,处死动物 ,观察组织病理学变化 .结果 :再灌注 2 0min后 ,与C组相比 ,H组TXB2 值明显降低 ,6 keto PGF1α明显增高 (P <0 .0 1) ;再灌注后 4 ,8,12 ,2 4及 4 8h时的神经功能评分H组明显高于C组 (P <0 .0 1) .光镜下 ,与C组相比 ,H组脊髓组织损伤显著减轻 ,形态基 p本正常的前角细胞较多 .结论 :高氧液对兔脊髓缺血再灌注损伤有显著的保护作用 .
AIM: To study the effects of hyperoxia liquid on spinal cord injury resulting from ischemia and reperfusion. METHODS: Twenty male New Zealand white rabbits were randomly divided into two groups: control group (Group C, n =10) and hyperoxia liquid group (Group H, n =10). The spinal cord ischemia rabbit model was made by occluding infrarenal abdominal aorta for 20 min. In Group H, hyperoxia liquid was administered intravenously 20 min before clamping at the rate of 20 ml·kg -1 ·h -1 and in Group C, saline was used in place of hyperoxia liquid. The levels of plasma TXB2 and the 6 keto PGF1α were assayed 20 min before ischmia, 10 min after ischmia and 20 min after reperfusion. Neurological functions were scored at 4, 8, 12, 24, 48 h after reperfusion. All animals were killed 48 h after reperfusion and the spinal cords were studied histopathologically. RESULTS: Twenty min after reperfusion, the level of plasma TXB2 in Group H was lower than that in Group C ( P < 0.01), but the level of plasma 6 keto PGF1α in Group H was higher than that in Group C ( P <0.01). Neurological function scores at 4, 8, 12, 24, 48 h after reperfusion were higher in Group H than those in Group C ( P <0.05). Compared with Group C, less degree of injury was seen in Group H under optical microscope 48h after reperfusion. CONCLUSION: Hyperoxia liquid can protect rabbit spinal cord injury resulting from ischemia and reperfusion.
