摘要
本实验应用杂交瘤技术建立两株抗人IgA单克隆抗体(McAbs),即AlA10和AlC9。此两株mcAbs针对人IgA的两个不同抗原决定簇,经等量混合后,在含有2%的聚乙二醇6000(PEG 6000)的1.5%琼脂糖免疫单扩散试验中,与多克隆抗体(PcAb)同时检测40份未经稀释的人血清样品,两者的相关性好(r=0.9493、P<0.001)。说明AlA10和AlC9混合后可替代PcAb在免疫单扩散中应用。
Among many McAbs to human IgA, two McAbs (A1A10 and A1C9) against two determinants were selected and mixed. They formed opaque precipitation rings with IgA in agarose containing 1% PEG 6000- SRID with our mixed McAbs or PcAbs was used to determine serum IgA concentration in 40 undiluted serum specimens: A comparison of the two showed similar results, with a correlation coefficient of 0.9493 (P<0.001).We conclude that mixed McAbs A1A10 and A1C9 can replace PcAbs in SRID for the measurement of human total serum IgA.
