人尿胰蛋白酶抑制剂工艺研究

IMPROVED PRODUCTION PROCESS OF URINE TRYPSIN INHIBITOR

报道了人尿胰蛋白酶抑制剂的提取新工艺。新鲜男性尿碱化除去沉淀,调 pH 4.0,用 PolyerdeⅣ搅拌吸附,吸附剂经抽滤、洗涤、NH_4HCO_3洗脱。冼脱液经硫酸铵沉淀、透析、冻干得产品。活性回收率为60～68%,比活为515～660 u/mg。

One hundred L of normal male human urine,freshly collected,were adjusted to pH 8.5 by addition of 1 mol/L NaOH.After 2h,the precipitare was removed,then the supernatant adjusted to pH 4.0 with lmol/L HCl.The solution,after removing of precipitate,was stirred with 500g of Polyerde Ⅳfor 2h.The Polyerde IV adsorbe(?)t was separated,washed with Water and eluted with NH_4HCO_3 solution,from which UTI was precipitated with(NH_4)_2SO_4 anddialyzed against phosphalate buffer solution and lyophilized.Specific activity ofthe UTI preparation was 515～660u/mg,with a recovery of 60～68%.
