摘要
利用改良的 Franz 扩散池,测定含不同促进剂的硝酸异山梨酯混悬液〔以甘油-丙二醇(1:1)作混悬介质〕的体外透皮作用,发现促进作用的大小顺序为5%尿素>3%月桂氮(艹卓)酮>5%二甲基亚砜。利用5%尿素为促进剂配制了涂膜剂,其透皮速度k=0.1396mg/cm^2·h;24h 累积透皮量为3.333±0.546mg/cm^2。
The effect of penetration enhancers on the in vitro permeabilityof isosorbide dinitrate(ID)from the suspension[glycerin-propylene glycol(1:1)as solvent]through rat skin was examined using the Franz's type diffusion cell and normal saline as recepter medium.The results showed that the penetrationrates of ID from 2.4%(W/V)suspension containing one of enhancers decreasedin following order:5%urea>3% laurolactone>5% dimethyl sulfoxide.A tran-sdermal therapeutic film of ID with 5% urea as enhancer was prepared.Thepermeability constant k was 0.1396mg/cm^2.h,while 24h accumulative quantityof penetration was 3.333±0.546mg/cm^2.
