摘要
目的 研究几种白血病和实体瘤细胞内三氧化二砷 (As2 O3 )浓度对其治疗敏感性的影响。方法 将几种白血病细胞株和原代细胞及实体瘤细胞株分别给予特定浓度和时间As2 O3 干预后 ,原子荧光法测定细胞内砷浓度。流式细胞仪检测细胞凋亡率。结果 一定浓度和时间As2 O3 干预后 ,不同细胞内药物浓度不同。NB4细胞株 >APL原代 >K5 6 2细胞株 >CML原代 >HL6 0细胞株 >AML M2原代 >HeLa细胞株 >肝癌腹水H2 2 细胞株 >胃腺癌SGC 790 1细胞株 >喉癌Hep 2细胞株。在最低促凋亡浓度水平以上的低浓度、长时间持续干预比短期高浓度干预的细胞内砷浓度高 ,凋亡率大。结论 细胞内砷浓度不同可能是As2 O3 治疗敏感性不同的原因之一。
OBJECTIVE: To study the relationship between intracellular concentrations of arsenic trioxide (As2 O3) in several kinds of leukemia or solid carcinoma cell strains and the therapeutic sensitivities. METHODS: Intracellular arsenic concentrations were assayed with atom fluorescent method. Apoptosis rate was evaluated with flow cytometry. RESULTS: After being managed by As2 O3 of different concentrations and for different durations, different cell strains had different intracellular arsenic concentrations. The sequence was as follows: NB4 cell line 〉 APL primary cell 〉 K562 cell line 〉 CML primary cell 〉 HL-60 cell line 〉 AML-M2 primary cell 〉 HeLa cell line 〉 H-22 cell 〉 SGC-7901 cell line 〉 cancer of larynx Hep-2 cell line. There were higher intracellular arsenic concentrations in NB4, K562 and APL cell strains cultured with As 2 O3 of lower concentrations above the lowest apoptotic concentration for a long time than As2 O3 of higher concentration for a short period. CONCLUSION: Different leukemic and solid carcinoma cell strains had different intracellular arsenic concentrations, which might be one of the factors that determined the sensitivities of leukemia and solid carcinoma to As2 O3 in clinical treatment.
