摘要
目的 观察川芎嗪对哮喘大鼠气道壁胶原合成的影响。方法 40只SD大鼠随机分成正常对照组、哮喘模型组、地塞米松组、大剂量和小剂量川芎嗪干预组。以卵蛋白 (OVA)致敏制备大鼠哮喘模型 ,肺组织石蜡切片行免疫组化染色计算机图像分析测定气道Ⅰ、Ⅲ型胶原含量 ,HE染色测气道壁内、外径及网状基底膜层厚度。结果 川芎嗪大剂量干预组与小剂量干预组气道壁内、外径比值均高于哮喘模型组 (P <0 0 0 1) ,网状基底膜厚度较哮喘模型组显著变薄 (P <0 0 0 1) ,与地塞米松组相似 ,川芎嗪两组间无明显差别 (P >0 0 5 )。川芎嗪两组气道壁Ⅲ型胶原含量显著低于哮喘模型组 (P <0 0 0 1) ,大剂量组作用强于小剂量组 (P <0 0 1)。结论 川芎嗪可以通过减少气道壁Ⅲ型胶原沉积和气道壁增厚而抑制气道重建。
Objective To observe the effect of Ligustrazine on the collagen deposition in asthmatic rats.Methods 40 SD rats were randomly divided into 5 groups:the normal group,the model group,Dexamethasone group,the low-dose and high-dose of Ligustrazing group,8 rats in each group.The changes of collagen content in the airway wall and the thickness of reticular basement membrane,inner and outer diameter were measured by the computerized image analysis system.Results The ratio of inner diameter to outer diameter in the low-dose and high-dose of Ligustrazing group were higher than that in the model group(P<0.001),and the wall contents of collagen type Ⅲ and the thickness of reticular basement membrane layer were significantly lower than that in the model group(P<0.001).The effect of the ligustrazine group were similar to that of dexamethasone group,and the high-dose of ligustrazine were preferable.Conclusion Ligustrazine could inhibit the airway remodeling by the way of reducing the precipitation of collagen and the thickness of airway wall.
