摘要
本文报道了用HPLC法对头孢哌酮(先锋必)在人体中药动学的研究以及血浆中药物浓度的测定。依次用乙腈、二氯甲烷沉淀血浆蛋白,取上清液直接进样。以ODS为固定相,甲醇:水:冰醋酸(300:700:1),用乙酸钠调pH至5.85为流动相,检测波长λ=254nm内标物为头孢拉定。头孢哌酮最低检出值1μg/ml,标准曲线y=1.2235x—0.005225,r=0.9998,线性范围2.5--800μg/ml,平均回收率为94.0±7.5%。本法对6名健康受试者体内药动学进行了研究,结果表明其处置过程符合线性二室模型,药时曲线方程C=173.20e^(-1.497t)+86.56e^(-0.2733t),人体内平均t_(1/2)β为2.6h。
Cefoperazone in human serum and it's pharmacokinetic parameters were determined by High-Performance Liquid Chro-matograph (HPLC). The protein in serum was precipitated, Using CH_3CN and CH_2Cl_2, the left liquid was directly taken forinjection. The solid phase was ODS and the mobile phase was a mixture of CH_3OH: H_2O: CH_3COOH=300: 700: 1 (pH ad-justed to 5.85 using CH_3COONa). The detection wavelength was 254nm. Cephradine was used as the internal standard. Dectec-tion limit was 1μg/ml. The linear equation was Y=1.2235X-0.005225, r=0.9998 and the linear range is 2.5~800μg/ml. The rate of average recovery was 94.0±7.5%. The pharmacokinetics of six health volunteers were studied. The resultshowed that handling process conformed to two-compartment open model. The linear equation of drug concentration in bloodand time was C=173.2e^(-1.497t)+86.56e^(-0.2733t). The average elimination half-life after intravenous injection was 2.60hours.
