静脉给药氧哌嗪青霉素的药物动力学

摘要 本文选取内科呼吸组住院病例10例,治疗量氧哌嗪青霉素4g静脉滴毕后用微量微生物法测定血药浓度,按双室模型计算结果。平均Cmax值为151.51±37.99μg/ml,T_(1/2)a为0.38±0.19h,T_(1/2)α。值为0.67±0.04h较文献报道1h短,给药后8h血药浓度趋于零。其它药动学参数Vc,Cl,AUC,Ke等值与文献报道相一致。本品4g/d分2次静滴治疗肺炎克雷白杆菌,变形,绿脓杆菌及伴厌氧菌致呼吸系统感染有效率达78%,副反应有胃肠反应及皮疹等。 10 cases of in-patients with bronchopulmonary infection were selected. Therapeutic dosage of pi-peracillin was given intravenously in each case there after, the elimination phase of serum drug concentra-tion was determined by biological assay, pharmacokinetic paremeters were calculated with the peak con-centration curve fitting using open 2-compartment model; average value of the peak concentration was151.51±37.99μg/ml, the main parameters were similar as those in the other reports. But the value oft_(1/2)b was less than one hour, 0.67±0.04 hour in this paper. Curative effect of piperacillin was better for bronchopulmonary infection with klebsilla, proteus vul-garis, pseudomonas agrugina saetc, side effects, such as G-I response and skin rash, were minimal.
