摘要
应用小鼠离体胰腺碎片灌流技术研究了在37、25、10和40℃下胰腺的淀粉酶释放以及乙酰胆碱(ACh)对胰淀粉酶释放的刺激效应。灌流液采用Krebs-Henseleit(KH)溶液,对照温度为37℃。实验观察到:25℃时淀粉酶的释放量减低,10℃时极明显减低,而40℃时淀粉酶释放量明显增加。在KH灌流液中加入ACh对淀粉酶释放有刺激效应。但是10℃时的刺激效应比37℃时大约降低一半,而40℃时ACh对淀粉酶的刺激效应仍保持较高水平。在离体条件下基础淀粉酶释放主要是在胰内神经节影响下神经末梢释放ACh的结果。结果表明,低温可降低胰腺泡细胞对ACh的敏感性。
The amylase release from mouse pancreatic fragments perfused with Krebs-Henseleit (KH) solution was studied at 37(control), 25, 10 and 40℃. The amylase release(U ·min-1·100mg-1 tissue) decreased at 25℃ and 10℃ in comparison with the basal amylase release, while increased at 40℃ . The amylase release from mouse pancreatic fragments was stimulated by addition of exogenous acetylcholine (ACh) into the perfusate, the stimulatory effect of which was diminished to about 50% at 10℃ as compared with that at control temperature. It is considered that the basal secretion in vitro is induced by ACh release from nerve endings under the influence of intrapancreatic ganglia and the sensitivity of pancreas acinar cell to ACh is depressed at low ambient temperature.
