抑癌基因PTEN在胰腺癌组织中的表达及其意义 被引量:2

摘要 目的 研究胰腺癌组织中PTEN蛋白及PTENmRNA的表达及其临床病理意义。方法 常规石蜡包埋切片SABC免疫组化法和原位杂交技术检测 2 6例胰腺癌、 12例癌旁组织及 8例正常胰腺组织中PTEN蛋白和PTENmRNA表达情况 ,同时结合病人的临床病理资料进行分析。结果  2 6例胰腺癌、 12例癌旁组织及 8例正常胰腺组织中 ,正常胰腺组织及癌旁组织PTEN蛋白阳性 16例 (75 % ) ,阳性物质位于胰腺腺细胞及胰岛细胞的胞质中 ,胰腺癌PTEN蛋白阳性 10例 ,PTEN蛋白表达于癌细胞胞质 ,阳性率 (38 4 % )与正常组织存在显著差异 (P <0 0 1)。淋巴结未转移病例PTEN阳性率与淋巴结转移病例的阳性表达率无显著差异 (P >0 0 5 )。PTENmRNA表达结果与PTEN蛋白基本一致 ,胰腺癌PTENmRNA阳性 12例 ,阳性率 (4 6 2 % )。结论 PTEN表达与胰腺癌临床病理特征和生物学行为存在密切关系。可能与胰腺癌的发生、发展及预后有关。 Objective To study the expression and clinical significance of PTEN protein and PTEN mRNA in the tissues of pancreatic cancer.Method SABC immunohistochemistry and in situ hybridization were used to detected the expression of PTEN protein and PTEN mRNA in 26 specimens of pancreatic cancer, 12 specimens of surrounding tissues and 8 specimens of normal pancreatic tissues, and the clinical pathologic data were analyzed.Result PTEN protein appeared mainly in the cytoplasm of pancreatic glandular cells and islet cells in 75% of the normal pancreatic tissues and the surrounding tissues, but in the cytoplasm of cancer cells in only 38.4% of the pancreatic cancer tissues. The difference in PTEN protein expression between normal and cancer tissues was significant (P<0.01). However, the difference in PTEN protein between specimens with lymph node metastasis and non-metastatic specimens was in significant (P>0.05). The expression of PTEN mRNA was consistent with that of the protein, being detected in 46.2% of pancreatic cancer tissues. Conclusion The expression of PTEN has a close relationship with the clinico-pathologic characters and biological behavior of pancreatic cancer, and may be associated with the occurrence, development and prognosis of the cancer.
