摘要
目的 探讨白细胞介素 1β (Interleukin 1beta ,IL 1β)对L 谷氨酸致痫大鼠大脑PLCβ1表达的影响 ,为阐明IL 1β在致痫中的作用机制提供资料。方法 随机将健康成年SD大鼠分为 5组 ,每组 8只 ,即对照组、Glu组、IL 1β +Glu组、IL 1ra +IL 1β +Glu组和MPEP +IL 1β +Glu组 ,采用行为学及Westernblot和免疫组织化学方法进行研究。结果 行为观察显示 ,对照组、IL 1ra +IL 1β +Glu组和MPEP组无痫性发作 ;Westernblot结果显示PLCβ1含量在Glu组和IL 1β +Glu组大鼠鼠脑皮质及海马内均较对照组、IL 1ra +IL 1β +Glu组和MPEP +IL 1β +Glu组明显增多 (P <0 0 5 ) ,而对照组、IL 1ra +IL 1β +Glu组和MPEP +IL 1β +Glu组组间无明显差别 (P >0 0 5 ) ;免疫组织化学染色显示 ,PLCβ1免疫反应增强主要表现在海马CA3区和大脑皮质 ,各组间的变化规律与Westernblot检测结果一致。结论 IL 1β对L 谷氨酸致痫有促进作用 ,mGluR5介导的PLCβ1激活参与了IL 1的促痫机制。
Objective In order to investigatethe mechanism of IL-1β in its facilitation of seizure,the influence of IL-1 on the expression of phospholipase Cβ1was studied in rats with seizure induced by L-glutamate. Method 40 healthy mature male SD rats were divided randomly into 5 groups: 1.Control group, 2.Glu group, 3. IL-1β+Glu group, 4.IL-1ra+IL-1β+Glu group, and 5.MPEP+IL-1β+Glu group. Behavior observation and immunohistochemicalstaining were used to assess the changes in the expression of phospholipase Cβ1in the brain. Result Thecontrol group, IL-1ra+IL-1β+Glu groupandMPEP+IL-1β+Glu group showed no seizure activities. Western blot showed that PLCβ1 wasincreased in the hippocampus area CA3 and the cerebral cortex of the Glu group and IL-1β+Glu group compared with that in the control group, IL-1ra+IL-1β+Glu group and MPEP+IL-1β+Glu group, but no significant variations were seen amongthe latter 3groups. Conclusion IL-1β plays a role in the facilitation of seizureinduced by L-glutamate, andthe activation of PLCβ1 by mGluR5 participates in the mechanism of IL-1 in facilitating seizure.
