摘要
目的 研究白细胞介素 1β (Interleukin 1β ,IL 1β)和谷氨酸 (Glutamicacid ,Glu)致痫过程中钙调神经磷酸酶 (Calcineurin ,CaN)在大脑皮质及海马内表达的变化 ,探讨IL 1在癫痫发病中的作用机制。方法 将实验大鼠随机分为3组 ,即 :A组 (生理盐水对照组 ) ,B组 (IL 1β组 ) ,C组 (IL 1β +Glu ,阈下剂量组 ) ,在大鼠行侧脑室注射相应试剂后30min、 6 0min观察大鼠行为变化并采用免疫组织化学方法检测大脑皮质及海马内CaN的表达。结果 行为观察结果 ,参照Schultz Krohn的评估标准A组无明显癫痫发作 ,B组发作程度达Ⅱ~Ⅲ级 ,C组达Ⅲ~Ⅳ级。免疫组化染色显示 ,在注射后30min ,B、C组CaN的表达无明显变化 ,6 0min明显升高 ,差异具有显著性意义。结论 IL 1β在促痫和致痫过程中通过某种途径缓慢激活CaN ,后者可能抑制癫痫发作。
Objective In order to investigatethe mechanism of IL-1β in its facilitation of seizure,changesof calcineurin immunoreaction was studied in the cerebral cortex and hippocampus of rats with seizure induced by L-glutamate and IL-1β. Method Experimental rats were dividedinto 3groups: group A (control group), group B (IL-1β group), and group C (IL-1β+ Glu group). After intracerebroventricular injection of reagents, the expression of calcineurin was examined by means of immunohistochemistry in the cerebral cortex and hippocampus at 30min and 60min. Result The results of the behaviour observation showed no seizure in group A,whileⅡ~Ⅲ grade sezure was observed in group B, andⅢ~Ⅳgrade in group C. The immunohistochemicalstaining showed that the expression of calcineurinwas significantly enhanced in groups B and C at 60min,but not at 30min after the injettion. Conclusion Calcineurin was slowly and gradually activated in the process of seizure induced by L-glutamate sodium and IL-1β, and it may inhibit the seizure.
